It depends on the aircraft you are flying, and whether you are taking off or landing. A Boeing 737 generally takes off with flaps set to 5 degrees, but can take off with them at 10 if necessary (almost never). On landing, the flaps are gradually extended as the aircraft slows until they are at 30 degrees. The 40 degree setting is generally only used for steep approaches, or to short runways.

Each aircraft has their own flap settings, but the example I provided will give you a good "rule of thumb," so to speak.
